General Procedure

1.Plasmid Preparation:

The target gene was amplified by PCR and inserted into the corresponding expression vector. The sequence of the constructed vector was validated by sequencing. The correct plasmid was amplified and transmitted to downstream.

2.Cell Culture and Transient Transfection:

For transient transfection, the plasmids were mixed with transfection reagents at an optimal ratio and then added into the flask or the bioreactor containing CHO. The cells were cultured in a serum-free medium and maintained in Erlenmeyer Flasks on an orbital shaker or the bioreactor by a suitable stirring speed at 37°C for 6 days.

3.Purification and Analysis:

1. Cell culture broth was centrifuged.
2. Cell culture supernatant was loaded onto affinity purification column at an appropriate flow rate.
3. The purified protein was analyzed by SDS-PAGE , SEC-HPLC and Elisa.
